This combination therapy consists of three agents: - **BMS-986288**, a probody therapeutic and modified version of ipilimumab (Yervoy), is an investigational anti-CTLA-4 antibody designed to be activated in the tumor microenvironment, minimizing systemic toxicity by masking its activity until proteolytically activated near tumors. - **Nivolumab** (Opdivo) is a fully human IgG4 monoclonal antibody that targets the programmed death-1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2, thereby enhancing T-cell mediated immune responses against cancer. - **Regorafenib** (Stivarga) is an orally administered multikinase inhibitor, inhibiting various kinases involved in tumor angiogenesis, oncogenesis, and maintenance of the tumor microenvironment, such as VEGFR, PDGFR, FGFR, KIT, RET, and RAF. The combination has been evaluated in advanced solid tumors, with primary investigation in metastatic colorectal cancer and advanced solid tumors refractory to standard care.
