Overview

BMS-986406 is a human IgG4 monoclonal antibody that targets the inhibitory immune checkpoint receptor leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2, also known as ILT4 or CD85d). By specifically binding to LILRB2 and blocking its interaction with ligands such as HLA-G, BMS-986406 is designed to modulate myeloid cell function and enhance antitumor immune responses. The drug is being developed by Bristol Myers Squibb for the treatment of advanced solid tumors, including metastatic renal cell carcinoma, metastatic hormone-refractory prostate cancer, metastatic ovarian cancer, melanoma, non-small cell lung cancer (including squamous subtype), pancreatic ductal adenocarcinoma, gastric cancer, recurrent head and neck squamous cell carcinoma, gastroesophageal junction carcinoma, bladder cancer, malignant mesothelioma, triple-negative breast cancer, and soft tissue sarcoma. As of early 2025 it remains in Phase 1 clinical trials[1][3][5][6][8].
