BMS-986416 is an investigational bispecific monoclonal antibody developed by Bristol Myers Squibb that simultaneously targets two inhibitory immune checkpoint receptors: T-cell immunoreceptor with Ig and ITIM domains (TIGIT) and Poliovirus receptor-related immunoglobulin domain-containing protein (PVRIG, also known as CD112R). Both TIGIT and PVRIG are expressed on T cells and natural killer (NK) cells and contribute to immune evasion by tumors. By blocking these pathways, BMS-986416 aims to restore and enhance anti-tumor immune responses. It is currently being evaluated in clinical trials for patients with advanced solid tumors, both as a monotherapy and in combination with other immunotherapies such as the PD-1 inhibitor nivolumab.
