Overview

BMS-986442 + nivolumab + carboplatin + paclitaxel is an **investigational combination regimen** composed of four agents: BMS-986442, a **bispecific monoclonal antibody** targeting TIGIT and CD96, with an enhanced Fc region; nivolumab, a **PD-1 immune checkpoint inhibitor** (marketed as Opdivo); carboplatin, a platinum-based **chemotherapy agent**; and paclitaxel, a **microtubule inhibitor** chemotherapy. The regimen is being evaluated primarily for the treatment of **non-small cell lung cancer (NSCLC)** and other solid tumors. BMS-986442 aims to boost immune system anti-tumor responses by dual blockade of TIGIT and CD96, augmenting T and NK cell activation and overcoming adaptive resistance, while nivolumab blocks PD-1 to restore T cell activity against tumors. Carboplatin and paclitaxel disrupt DNA replication and microtubule function, respectively, leading to direct tumor cytotoxicity. The regimen was pursued in a Phase 1/2 study, but the chemotherapy combination with BMS-986442 and nivolumab was not initiated due to early trial termination for business reasons, and not for safety concerns[1][3][4][5].
