Overview

BMS-986449 is an investigational oral molecular glue degrader developed by Bristol Myers Squibb for the treatment of advanced solid tumors. It functions as a cereblon E3 ligase modulatory drug (CELMoD), selectively targeting and degrading the transcription factors IKZF2 (Helios) and IKZF4 (Eos), which are members of the Ikaros family of zinc finger proteins. By promoting proteasomal degradation of these proteins via cereblon engagement, BMS-986449 modulates immune cell function and may enhance anti-tumor immunity. The drug is being evaluated both as monotherapy and in combination with nivolumab in clinical trials for advanced solid malignancies, including non-small cell lung cancer (NSCLC) and triple-negative breast cancer (TNBC)[2][3][5].
