Overview

**BMS-986458 + rituximab** is a combination therapy under investigation in relapsed/refractory non-Hodgkin lymphoma (NHL), especially diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma (FL). BMS-986458 is an orally bioavailable, selective ligand-directed degrader of B-cell lymphoma 6 protein (BCL6) that acts via cereblon-dependent E3 ubiquitin ligase-mediated protein degradation, resulting in reduced expression of BCL6 and potent antitumor activity. This degradation increases CD20 surface expression, potentially sensitizing lymphoma cells to rituximab, a monoclonal antibody targeting CD20 that mediates B-cell lysis primarily through antibody-dependent cellular cytotoxicity (ADCC) and complement activation. The combination is being evaluated in early-phase clinical trials for its safety, tolerability, pharmacokinetics, and preliminary efficacy in patients whose disease has relapsed or is refractory to standard therapies[1][2][3][4][7].
