BMS-986482 + nivolumab + relatlimab + rHuPH20 is an investigational multi-agent combination therapy being studied for advanced solid tumors. BMS-986482 is a small molecule in development by Bristol Myers Squibb, likely representing a novel immunomodulator. Nivolumab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1 and PD-L2, thus inhibiting immune checkpoint-mediated immune evasion by tumors. Relatlimab is a monoclonal antibody targeting lymphocyte-activation gene 3 (LAG-3), another immune checkpoint receptor predominantly on T cells, blocking its immunosuppressive function. Opdualag, an FDA-approved fixed-dose combination of nivolumab and relatlimab, is used for unresectable/metastatic melanoma. rHuPH20 (recombinant human hyaluronidase PH20, developed by Halozyme) is a recombinant enzyme that transiently degrades hyaluronan to enhance subcutaneous drug absorption and dispersion, facilitating administration of large volume subcutaneous injections. The combination is under investigation in a Phase 1/2 trial (NCT06697197) for advanced solid tumors.
