BMS-986497 (formerly ORM-6151) is an investigational anti-CD33 antibody neoDegrader conjugate (AnDC) developed for the treatment of relapsed or refractory acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS). The drug consists of an antibody that specifically targets CD33-expressing tumor cells and delivers a molecular glue degrader payload that induces degradation of the peptide-chain-release factor 3 protein GSPT1 within cancer cells. This targeted degradation leads to apoptosis in both dividing and non-dividing malignant cells, offering a novel mechanism distinct from traditional DNA-damaging chemotherapies. The drug is currently being evaluated in Phase 1 clinical trials to assess its safety, tolerability, pharmacokinetics, efficacy, and recommended dosing in adults with relapsed or refractory AML or MDS[2][4][6].
