Overview

BMS-986500 is an investigational small molecule drug being developed by Bristol Myers Squibb for the treatment of advanced solid tumors, including advanced breast cancer and ovarian cancer. It is currently in phase 1 clinical trials to assess its safety, tolerability, and initial efficacy, both as monotherapy and in combination with other agents in patients who have previously been treated with CDK4/6 inhibitors. The exact mechanism of action has not been publicly disclosed. However, based on the clinical trial design focusing on CDK4/6 inhibitor-pretreated breast cancer and CCNE1-amplified ovarian cancer, it is hypothesized that BMS-986500 may target cyclin-dependent kinases (CDKs), possibly aiming to overcome acquired resistance to CDK4/6 inhibition. Another speculation in the oncology field is potential PKMYT1 inhibition, which represents a synthetic lethality approach, although there is no confirmed data on this[1][2][5][7][9][10].
