Overview

BN-Brachyury is a cancer immunotherapy vaccine developed by Bavarian Nordic in collaboration with the National Cancer Institute for the treatment of advanced chordoma. The vaccine utilizes a prime-boost regimen consisting of two viral vector platforms: modified vaccinia Ankara-Bavarian Nordic (MVA-BN)-Brachyury as the priming vaccine and fowlpox virus (FPV)-Brachyury as the boosting vaccine. Both vectors are engineered to encode the brachyury gene—a transcription factor universally expressed in chordoma but rare in normal tissues—alongside three costimulatory molecules (B7.1, LFA-3, and ICAM-1, collectively known as TRICOM). This combination is designed to activate the immune system to recognize and destroy brachyury-expressing tumor cells. In clinical trials, BN-Brachyury is administered subcutaneously in combination with radiation therapy, which may act synergistically to enhance the anti-tumor immune response. The FDA granted orphan drug designation to BN-Brachyury for chordoma in 2018.
