BN-CD38 is a compact, single-chain CD38–CD3 T‑cell engager antibody construct designed to redirect autologous T cells against CD38-positive acute myeloid leukemia (AML) blasts and leukemia stem cells. By simultaneously binding CD38 on AML cells and CD3 on CD4 and CD8 T cells, BN-CD38 promotes formation of an effective immunological synapse, triggering T-cell activation, proliferation, and cytotoxicity against leukemia cells.[2][13] BN-CD38–induced interferon-gamma release upregulates CD38 on otherwise CD34+CD38− leukemia stem cell–enriched blasts, thereby enabling their subsequent targeting while sparing normal hematopoietic stem cells in preclinical models.[2][13] This investigational biologic has been characterized preclinically for AML and leukemia stem cell eradication but does not yet have reported clinical development data.[2][11][13]
