Overview

BN-CD38mut is a mutant, non-binding control version of the therapeutic candidate BN-CD38. BN-CD38 is a compact, single-chain T-cell engager designed to target CD38-expressing cancer cells and CD3-expressing T cells. BN-CD38mut is specifically engineered to lack binding affinity for CD38, serving as a negative control in preclinical studies for acute myeloid leukemia (AML) and multiple myeloma (MM). It was developed by researchers at the City of Hope to validate the target-specific activity of the parent BN-CD38 molecule. In experimental models, BN-CD38mut shows minimal T-cell activation and significantly lower cytotoxicity compared to the active BN-CD38 construct, which utilizes a CD38 nanobody inserted between the light and heavy chains of an anti-CD3 Fab.
