BNC101 is a humanized monoclonal antibody that targets leucine-rich repeat-containing G-protein coupled receptor 5 (LGR5), a marker of cancer stem cells. By binding to LGR5, BNC101 inhibits the growth and survival of cancer stem cells, which are implicated in tumor initiation, recurrence, and metastasis. The drug was developed primarily for the treatment of metastatic colorectal cancer and other solid tumors where LGR5 is overexpressed. Preclinical and early clinical data suggested that targeting LGR5 could prevent or delay tumor recurrence by eliminating these therapy-resistant cell populations[2][3][6]. The drug was administered intravenously as monotherapy or in combination with chemotherapy[1][5]. Despite initial promise and evidence of target engagement in Phase 1 trials, development for all indications has been discontinued[3].
