BNC105 is a small molecule vascular disrupting agent (VDA) and tubulin polymerization inhibitor developed for the treatment of solid tumors. Its primary mechanism involves selectively targeting and disrupting tumor vasculature by inhibiting tubulin polymerization in proliferating endothelial cells within tumors. This leads to occlusion of tumor blood vessels and subsequent deprivation of oxygen and nutrients to cancer cells. In addition to its anti-vascular effects, BNC105 exerts direct cytotoxicity on tumor cells through mitotic spindle disruption and cell cycle arrest. The compound demonstrates high selectivity for activated endothelial cells over normal vasculature and is rapidly cleared from normal tissues but retained in tumors—a property that enhances its therapeutic index while minimizing systemic toxicity[1][2][3][4][5]. Developed using proprietary MultiCore technology by Bionomics (now a subsidiary of Neuphoria Therapeutics), it has been evaluated as monotherapy or in combination with standard chemotherapies such as carboplatin/gemcitabine or mTOR inhibitors[1][2][8]. Clinical trials have shown promising activity in ovarian cancer and renal cell carcinoma subgroups[6][8].
