BND-67 is a first-in-class single-domain VHH antibody (nanobody) developed by Biond Biologics. It is rationally designed to bind the shedding-prone region of the CD28 receptor on T cells and sterically hinder matrix metalloproteinases (MMPs) from cleaving CD28. This prevents the release of soluble CD28, which acts as a decoy receptor and suppresses T cell effector function, thereby attenuating anti-PD1 therapy effects. By inhibiting CD28 shedding without agonistic or antagonistic activity on the receptor itself, BND-67 aims to increase membranal CD28 density on T cells, augmenting their activation and potentially overcoming resistance to anti-PD1 immunotherapy in cancer patients[1][2][3][8].
