BnoX is a sublingual buprenorphine oral wafer developed by iX Biopharma using its proprietary WaferiX drug delivery technology. It is designed for the treatment of acute and chronic pain, as well as opioid use disorder. The formulation consists of a rapidly dissolving wafer that delivers buprenorphine, a partial mu-opioid receptor agonist and kappa-opioid receptor antagonist, directly into the sublingual mucosa. This delivery method bypasses first-pass metabolism, resulting in improved bioavailability (45.4%) and a faster onset of action (median Tmax of 1.0 hour) compared to traditional sublingual tablets like Temgesic. BnoX leverages buprenorphine's ceiling effect on respiratory depression and its long duration of action to provide a safer and more effective analgesic profile.
