Overview

BNT112 is an investigational mRNA-based cancer vaccine developed by BioNTech. It encodes a fixed set of five prostate cancer-associated antigens designed to trigger a strong and precise immune response against prostate tumors. The vaccine is formulated as a uridine mRNA-lipoplex and aims to stimulate both CD4+ and CD8+ T cell responses by presenting tumor-specific antigens to the immune system. BNT112 has been evaluated in Phase 1/2 clinical trials (PRO-MERIT) as monotherapy and in combination with cemiplimab (a PD-1 inhibitor) for patients with metastatic castration-resistant prostate cancer (mCRPC) and high-risk localized prostate cancer. Initial data indicated robust antigen-specific immune responses and an acceptable safety profile; however, recent trials have been terminated due to sponsor decision[1][2][3][4].
