BNT116 + ALK inhibitor + RET inhibitor is a combination therapy regimen developed by BioNTech SE for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC). The regimen centers on BNT116, an intravenously administered, unmodified mRNA-based lipoplex cancer vaccine that encodes six tumor-associated antigens frequently expressed in NSCLC: CLDN6, KK-LC-1, MAGE-A3, MAGE-A4, MAGE-C1, and PRAME. In the LuCa-MERIT-1 Phase I trial (NCT05142189), BNT116 is being evaluated in combination with oral small-molecule tyrosine kinase inhibitors (TKIs) targeting either anaplastic lymphoma kinase (ALK) or rearranged during transfection (RET) in patients whose tumors harbor these specific genetic rearrangements. The vaccine is designed to stimulate a robust T-cell response against cancer cells, potentially enhancing the clinical benefit of targeted TKI therapy.
