BNT116 + anti-B7-H3 antibody conjugated to topoisomerase I inhibitor is an investigational combination therapy currently under clinical evaluation for advanced non-small cell lung cancer (NSCLC). BNT116 is an mRNA-based immunotherapy developed by BioNTech, designed as a cancer vaccine to elicit immune responses against selected tumor antigens. The anti-B7-H3 antibody conjugated to a topoisomerase I inhibitor is an antibody-drug conjugate (ADC) targeting B7-H3 (CD276), an immune checkpoint molecule frequently overexpressed in malignant solid tumors and associated with immune evasion and tumor progression. The ADC consists of a human or humanized anti-B7-H3 monoclonal antibody linked via a cleavable linker to a topoisomerase I inhibitor payload. The ADC binds to B7-H3 on tumor cells, facilitating internalization and release of the cytotoxic payload to induce tumor cell death. This combination aims to enhance antitumor immune responses and directly kill tumor cells. The primary indication under investigation is NSCLC[2][3][4].
