BNT116 + docetaxel is a combination therapy being investigated for non-small cell lung cancer (NSCLC). BNT116 is an mRNA-based cancer vaccine developed by BioNTech that targets six tumor-associated antigens commonly expressed in NSCLC, while docetaxel is a standard chemotherapy drug. The combination has shown encouraging preliminary results in clinical trials, with data presented at the AACR Annual Meeting in April 2024 indicating an overall response rate of 30% and a disease control rate of 85% in patients who had previously progressed on PD-1/PD-L1 inhibitors and platinum-based chemotherapy[5]. The combination demonstrated a manageable safety profile with no signs of additive toxicity[5]. ## Mechanism of Action BNT116 is an intravenously administered RNA-lipoplex therapeutic cancer vaccine comprising six RNAs encoding tumor-associated antigens (MAGE A3, CLDN6, KK-LC-1, PRAME, MAGE A4, and MAGE C1) that are frequently expressed in NSCLC[6]. The vaccine aims to stimulate a precise immune response against tumor cells by presenting these common tumor markers to the patient's immune system[8][10]. Docetaxel is a conventional chemotherapy drug that works by disrupting cell division. The combination therapy appears to have synergistic anti-tumor effects[5][9]. ## Clinical Development The combination is currently being evaluated in the LuCa-MERIT-1 trial (NCT05142189), a first-in-human Phase I trial designed to establish the safety profile and determine a safe dose for BNT116 alone and in various combinations[1][2]. The trial is recruiting patients across 34 research sites in seven countries, including the UK, US, Germany, Hungary, Poland, Spain, and Turkey[8][10]. Preliminary data has shown encouraging antitumor activity, consistent induction of immune responses, and a manageable safety profile without signs of additive toxicity[5].
