BNT141 is an investigational mRNA-based drug developed by BioNTech for the treatment of solid tumors expressing Claudin 18.2 (CLDN18.2). It consists of a lipid nanoparticle (LNP) formulation encapsulating nucleoside-modified messenger RNA that encodes for a monoclonal IgG1 antibody sequence-identical to IMAB362/Zolbetuximab, which targets the tumor-associated antigen CLDN18.2. Upon intravenous administration, the LNPs are taken up primarily by hepatocytes, where they release the mRNA into cells to produce and secrete the functional anti-Claudin18.2 antibody in vivo[1][5][7]. The mechanism of action involves binding to CLDN18.2 on tumor cells, leading to immune-mediated cytotoxicity and antitumor effects[1][3][5]. Preclinical studies have shown that BNT141 mediates cytotoxicity comparable to recombinant IMAB362 protein and inhibits growth of human tumor xenografts in animal models without significant toxicity[5][7]. Clinical development has focused on advanced or metastatic solid tumors positive for CLDN18.2 expression, including gastric, pancreatic, ovarian, and biliary tract cancers[4].
