Overview

BNT141 + nab-paclitaxel + gemcitabine is an investigational combination therapy currently being evaluated primarily for advanced CLDN18.2-positive solid tumors, especially pancreatic adenocarcinoma and cholangiocarcinoma. BNT141 is a nucleoside-modified mRNA therapeutic formulated in lipid nanoparticles that encodes the anti-Claudin 18.2 monoclonal antibody sequence (identical to IMAB362/Zolbetuximab). It is delivered via mRNA/LNP technology to induce in vivo production of a CLDN18.2-targeting antibody, which mediates direct antitumor cytotoxicity and antibody-dependent cellular cytotoxicity (ADCC). Nab-paclitaxel is a nanoparticle, albumin-bound formulation of the taxane chemotherapeutic paclitaxel, enhancing tumor delivery through albumin-mediated transport and reduced toxicity compared to solvent-based paclitaxel. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, serving as a backbone chemotherapeutic in pancreatic cancer. This combination is being investigated in Phase I/II trials, both in dose-escalation (to define maximum tolerated dose and recommended Phase II dose) and expansion cohorts, for patients with advanced CLDN18.2+ tumors who are eligible for nab-paclitaxel/gemcitabine therapy[1][3][5].
