BNT142 is an investigational mRNA-based drug developed by BioNTech for the treatment of advanced solid tumors that express Claudin 6 (CLDN6), an oncofetal antigen absent from normal adult tissues but present in various cancers including testicular, ovarian, endometrial, and non-squamous non-small cell lung cancer. The drug consists of a lipid nanoparticle-formulated RNA encoding a bispecific antibody that binds to both CD3 on T cells and CLDN6 on tumor cells. After intravenous administration, the RNA is translated in vivo into the functional bispecific antibody (RiboMab02.1), which then engages T cells to recognize and destroy CLDN6-positive cancer cells. Preclinical studies demonstrated potent antitumor activity with favorable safety profiles in animal models. BNT142 is currently being evaluated in Phase I/II clinical trials for safety and preliminary efficacy in patients with CLDN6-positive advanced solid tumors[1][2][4][5][6][7][8].
