Overview

BNT162b4 is an investigational mRNA-based vaccine developed to enhance T cell immunity against SARS-CoV-2. Unlike the widely used spike-protein-targeting vaccines, BNT162b4 encodes variant-conserved immunogenic segments of the SARS-CoV-2 nucleocapsid (N), membrane (M), and ORF1ab proteins. This design aims to broaden immune responses by targeting viral regions less prone to mutation across variants. The vaccine elicits robust polyfunctional CD4+ and CD8+ T cell responses in preclinical models and is intended for use alone or in combination with spike-based vaccines such as BNT162b2. Its primary indication is for COVID-19 prevention, with a focus on providing protection against severe disease from diverse SARS-CoV-2 variants[1][2][7].
