BNT162b4 + BNT162b2 is an investigational combination mRNA vaccine developed by BioNTech and Pfizer for the prevention of COVID-19. This combination therapy pairs BNT162b2 (Comirnaty), a nucleoside-modified mRNA vaccine encoding the SARS-CoV-2 spike protein, with BNT162b4, a next-generation vaccine candidate encoding conserved non-spike proteins including the nucleocapsid (N), membrane (M), and ORF1ab proteins. While BNT162b2 primarily induces neutralizing antibodies against the spike protein, BNT162b4 is designed to enhance and broaden T-cell mediated immunity by targeting viral proteins that are more conserved across different SARS-CoV-2 variants. The combination is being evaluated as a booster to provide more durable and comprehensive protection against evolving strains of the virus.
