BNT211 is an investigational autologous chimeric antigen receptor (CAR)-T cell therapy targeting the oncofetal antigen Claudin 6 (CLDN6), which is highly and specifically expressed in various solid tumors but not in healthy adult tissues. The product consists of patient-derived T cells genetically engineered to express a CAR specific for CLDN6. In clinical trials, BNT211 has been evaluated both as monotherapy and in combination with a nanoparticulate RNA vaccine (CARVac) designed to amplify and expand the transferred CAR-T cells in vivo. The primary indications under investigation are relapsed or refractory CLDN6-positive solid tumors—including testicular cancer and extragonadal germ cell tumors—after prior salvage therapies. Early-phase studies have shown promising anti-tumor activity with manageable toxicity profiles[1][4][8][10].
