Drug intelligence / Profile preview
BNT3210 is an investigational antibody-drug conjugate (ADC) developed by BioNTech that targets the tumor-associated carbohydrate antigen Sialyl-Tn (STn). Sialyl-Tn is a truncated O-glycan structure overexpressed in various epithelial cancers and solid tumors, while being virtually absent in healthy tissues. BNT3210 consists of a humanized IgG1 monoclonal antibody specific for Sialyl-Tn conjugated to a topoisomerase 1 inhibitor payload, specifically an exatecan derivative (DXd), via a cleavable linker. Upon binding to Sialyl-Tn on the surface of cancer cells, the ADC is internalized, releasing the cytotoxic payload to inhibit DNA topoisomerase 1, leading to DNA damage and cell death. BNT3210 is currently being evaluated in a Phase I/II clinical trial for the treatment of advanced solid tumors expressing Sialyl-Tn.
