BNT327 + topotecan is a **combination therapy** under clinical evaluation for the treatment of small-cell lung cancer (SCLC), particularly in patients whose disease has progressed after first- or second-line treatment. **BNT327 (also called pumitamig)** is an investigational bispecific antibody targeting **PD-L1** and **VEGF-A**, developed by BioNTech as a pan-tumor platform for combination therapies. **Topotecan** is a well-established chemotherapy agent that inhibits **topoisomerase I**, thereby preventing DNA re-ligation and resulting in cancer cell death. The combination aims to target immune evasion (PD-L1), block tumor angiogenesis (VEGF-A), and induce direct cytotoxicity (topotecan), representing a multi-modal strategy for resistant cancers. The regimen is currently being assessed for safety, efficacy, and dosing in several active Phase II/III clinical trials.
