BNT411 is a selective, intravenously administered small molecule agonist of Toll-like receptor 7 (TLR7), developed by BioNTech for the treatment of solid tumors, including extensive-stage small cell lung cancer (ES-SCLC). As an imidazoquinoline-based TLR7 agonist, BNT411 activates TLR7 signaling pathways, leading to broad innate and adaptive immune responses. This includes activation of dendritic cells, monocytes, natural killer cells, T cells, and B cells as well as systemic release of type I interferons and proinflammatory cytokines. Preclinical and early clinical studies have shown that BNT411 can induce immune modulation with antitumor activity signals in patients with various solid cancers. It is being investigated both as a monotherapy and in combination with cytotoxic therapies or immune checkpoint inhibitors.
