Overview

BO-1393 is a preclinical-stage N-mustard-quinoline conjugate bearing a hydrazinecarboxamide linker, designed as a DNA-directed alkylating agent. Developed by researchers at Academia Sinica and National Yang-Ming University in Taiwan, BO-1393 is synthesized by linking an alkylating nitrogen mustard warhead to a DNA-affinic quinoline carrier. This design enhances DNA-specific cross-linking while reducing the chemical reactivity and toxicity typically associated with nitrogen mustards. In preclinical animal models, BO-1393 has demonstrated significant antitumor activity, inducing complete remission in human breast carcinoma MX-1 xenografts, suppressing human ovarian SK-OV3 xenografts, and prolonging survival in mice bearing orthotopically implanted human glioma SK-NA-S xenografts.
