BO-212 is a next-generation systemic immunotherapy candidate developed by Highlight Therapeutics. It is a double-stranded RNA (dsRNA) complex designed to act as a potent agonist of Toll-like receptor 3 (TLR3), Retinoic acid-inducible gene I (RIG-I), and Melanoma Differentiation-Associated protein 5 (MDA5). While the company's lead candidate, BO-112, is restricted to intratumoral administration, BO-212 is optimized for systemic (intravenous) delivery. This allows the drug to target metastatic lesions and tumors that are not easily accessible for direct injection. By mimicking a viral infection, BO-212 activates the innate immune system within the tumor microenvironment, promoting the release of pro-inflammatory cytokines and enhancing the presentation of tumor antigens to prime a systemic T-cell-mediated anti-tumor response.
