BO-2329 is a novel bis(hydroxymethyl)indolizino[8,7-b]indole hybrid developed by researchers at the Institute of Biomedical Science, Academia Sinica. It is designed as an antitumor agent specifically targeting small cell lung cancer (SCLC). The molecule is a hybrid of β-carboline and bis(hydroxymethyl)pyrrole moieties. Its mechanism of action involves the induction of DNA cross-linking and inhibition of topoisomerase II (Topo II). In preclinical studies, BO-2329 demonstrated potent growth inhibition in SCLC cell lines (such as H526 and H211), induced cell cycle arrest at the G2/M phase, and triggered apoptosis. Structurally, the presence of an N11-methyl group in BO-2329 favors DNA cross-linking activity over Topo II inhibition compared to its N11-H analog, BO-2239.
