BODIPY650-cobalamin is a light- and X-ray-activated theranostic agent designed for targeted imaging and drug delivery in oncology. It consists of a cobalamin (vitamin B12) scaffold conjugated to the fluorophore BODIPY 650. The compound utilizes the transcobalamin receptor (TCblR/CD320) for cellular uptake, a receptor that is significantly overexpressed in various malignancies, including pancreatic adenocarcinoma (PDAC). The alkylcobalamin linkage is sensitive to both red light (650 nm) and clinical X-ray doses, allowing for the controlled release of cargo. This dual-trigger mechanism enables deep-tissue activation and potential synergy with standard-of-care radiotherapy, facilitating both the visualization of tumors and the localized release of therapeutic agents.
