Bomulrelvir (MK-1289) is an investigational, orally bioavailable small molecule inhibitor of the SARS-CoV-2 3C-like protease (3CLpro), also known as the main protease (Mpro). Developed by Merck & Co., it is designed for the treatment of COVID-19. The 3CLpro enzyme is essential for the viral life cycle as it cleaves the large polyproteins translated from the viral RNA into functional non-structural proteins required for replication. By inhibiting this enzyme, bomulrelvir prevents viral maturation and proliferation. Unlike some other 3CLpro inhibitors, bomulrelvir is being developed as a standalone agent that does not require co-administration with a pharmacokinetic booster like ritonavir, potentially reducing the risk of drug-drug interactions. It is currently in Phase 3 clinical development for non-hospitalized patients with COVID-19.
