Bone Morphogenetic Protein 4 (BMP4) is a signaling protein belonging to the TGF-β superfamily that plays a pivotal role in embryonic development, including bone formation (osteogenesis), neurogenesis, and vascular development. As a therapeutic candidate, it is being explored in various modalities, including recombinant protein and gene therapy (e.g., AAV8-BMP4), for its ability to modulate cell growth and differentiation. In oncology, BMP4 has shown potential in inducing growth arrest and apoptosis in multiple myeloma and pancreatic cancer cells, while in regenerative medicine, it is investigated for promoting osteogenic differentiation in bone defects and nonunions. Its mechanism of action involves binding to type I and type II serine/threonine kinase BMP receptors, which subsequently activates the canonical SMAD1/5/8 signaling pathway.
