Overview

Bone morphogenetic protein 5 (BMP-5) is a recombinant protein and a member of the transforming growth factor-beta (TGF-β) superfamily. It is a pleiotropic ligand known for its critical role in inducing bone and cartilage formation during development. BMP-5 initiates signaling by binding to the type I receptor BMPR1A and the type II receptor BMPR2, which activates the canonical SMAD signaling pathway. It is also capable of triggering non-canonical pathways, such as MAPK/p38 signaling. Research has explored the role of BMP-5 in various conditions, including osteoarthritis, vascular and valvular homeostasis, and certain cancers like pancreatic cancer, where it has shown bidirectional effects on cell growth and invasion. Currently, BMP-5 is primarily utilized as a research-use-only (RUO) reagent and does not have an identified clinical-stage developer or approved therapeutic indication.
