Bone morphogenetic protein 6 (BMP6) is a member of the transforming growth factor-beta (TGF-β) superfamily of signaling molecules. It plays a pivotal role in bone metabolism and is the primary endogenous regulator of systemic iron homeostasis through the induction of hepcidin. In the context of bone disease, BMP6 promotes the differentiation of mesenchymal stromal cells (MSCs) into osteoblasts, effectively restoring bone formation that is typically suppressed in conditions like multiple myeloma. Furthermore, BMP6 has demonstrated the ability to directly inhibit the proliferation of certain tumor cells, such as plasma cell malignancies, and attenuate inflammatory phenotypes in the bone marrow microenvironment. As a therapeutic agent, recombinant BMP6 is being investigated for its potential to treat bone loss, iron overload disorders, and specific cancers.
