Bordetella pertussis whole cell vaccine is an inactivated bacterial vaccine traditionally used for the prevention of pertussis (whooping cough). In the context of oncology, it is being investigated as a potent immunological adjuvant due to its high content of pathogen-associated molecular patterns (PAMPs), such as lipopolysaccharide (LPS) and lipoproteins, which act as ligands for various Toll-like receptors (TLRs). By stimulating TLRs (primarily TLR2 and TLR4), the vaccine promotes the maturation of dendritic cells and the secretion of pro-inflammatory cytokines, thereby enhancing the cellular immune response against co-administered tumor-associated antigens. NEC Corporation is evaluating this vaccine in combination with a WT1 peptide (W10) for the treatment of castration-resistant prostate cancer in Japan, aiming to leverage its adjuvant properties to stimulate antitumor immunity.
