This is a combination chemotherapy regimen consisting of three agents: - **Bortezomib** (a proteasome inhibitor), - **Rituximab** (a monoclonal antibody targeting CD20 on B cells), - **Bendamustine** (an alkylating agent with antimetabolite properties). The combination has been studied primarily for the treatment of relapsed or refractory indolent and mantle cell non-Hodgkin lymphoma. Laboratory studies suggest synergistic activity among these drugs. Clinical trials have demonstrated high response rates and manageable toxicity in heavily pretreated patient populations. The mechanism involves direct cytotoxicity to malignant lymphocytes via DNA damage (bendamustine), inhibition of protein degradation pathways leading to apoptosis (bortezomib), and immune-mediated destruction of CD20-positive B cells (rituximab)[1][2][3].
