bortezomib + cyclophosphamide + thalidomide + dexamethasone refers to the collective set of therapeutic agents utilized in a Phase 3 clinical trial (NCT01002183) conducted by the Grupo de Estudos Multicentricos em Onco-Hematologia (GEMOH). This multicenter Brazilian study compared two distinct triple-drug induction regimens for transplant-eligible patients with newly diagnosed multiple myeloma: VCD (comprising bortezomib, cyclophosphamide, and dexamethasone) and CTD (comprising cyclophosphamide, thalidomide, and dexamethasone). Bortezomib acts as a proteasome inhibitor, cyclophosphamide as an alkylating agent, thalidomide as an immunomodulatory drug targeting cereblon, and dexamethasone as a corticosteroid. The trial aimed to determine the superior induction protocol, ultimately finding that the bortezomib-containing VCD regimen achieved significantly higher response rates than the thalidomide-containing CTD regimen.
