This is a combination chemotherapy regimen consisting of the proteasome inhibitor bortezomib, the corticosteroid dexamethasone, and the alkylating agent cyclophosphamide. It is being evaluated by The First Affiliated Hospital of Soochow University for the treatment of Waldenström macroglobulinemia, a rare B-cell lymphoproliferative disorder. The regimen, often abbreviated as BCD or PCD, works by targeting the 26S proteasome to induce apoptosis in malignant cells, while cyclophosphamide cross-links DNA to inhibit cell division, and dexamethasone provides anti-inflammatory and lympholytic effects. A Phase 1/2 trial (NCT02971982) compared this regimen against the RCD (rituximab, cyclophosphamide, dexamethasone) protocol as induction and maintenance therapy.
