Overview

Bortezomib + docetaxel is a combination regimen of two small molecule chemotherapeutic agents used in the treatment of various solid tumors, particularly in patients with refractory or recurrent disease. Bortezomib is a first-in-class proteasome inhibitor that disrupts the 26S proteasome, leading to inhibition of protein degradation and induction of apoptosis in cancer cells. Docetaxel is a taxane that stabilizes microtubules and inhibits their depolymerization, resulting in cell cycle arrest and apoptosis. Preclinical studies have shown synergistic antitumor activity when these drugs are combined. The combination has been evaluated primarily for advanced solid tumors such as prostate cancer and other refractory malignancies[1][2][3].
