Overview
Bortezomib + IGEV is a combination chemotherapy regimen used primarily in the treatment of relapsed or refractory Hodgkin lymphoma. The regimen consists of bortezomib (a proteasome inhibitor) added to the standard IGEV protocol, which includes ifosfamide, gemcitabine, vinorelbine, and prednisone. Bortezomib acts by reversibly inhibiting the 26S proteasome in cells, leading to cell cycle arrest and apoptosis of cancer cells. The addition of bortezomib to IGEV has been studied for its potential to improve outcomes prior to high-dose chemotherapy and autologous stem cell transplantation in patients with Hodgkin lymphoma who have not responded adequately to previous treatments[1][2]. Clinical studies indicate that while toxicity is similar between regimens with or without bortezomib, adding it does not significantly improve response rates but maintains a favorable safety profile[2].
