Bortezomib + pegylated liposomal doxorubicin is a combination regimen used primarily in the treatment of multiple myeloma, especially in patients who have received at least one prior therapy. Bortezomib is a small molecule proteasome inhibitor that disrupts the 26S proteasome, leading to apoptosis of cancer cells. Pegylated liposomal doxorubicin is an anthracycline antibiotic encapsulated in PEG-coated liposomes, which allows for prolonged circulation and targeted delivery to tumor tissue; it acts by intercalating DNA and inhibiting nucleic acid synthesis. The combination has demonstrated improved time to disease progression compared with bortezomib alone in relapsed or refractory multiple myeloma and is FDA approved for this indication. It has also been studied in metastatic breast cancer but showed minimal activity in heavily pretreated populations[1][2][5][6].
