A combination therapy consisting of bortezomib, a proteasome inhibitor, and pemetrexed, an antifolate antineoplastic agent. Bortezomib works by reversibly inhibiting the 26S proteasome, leading to cell cycle arrest and apoptosis of cancer cells. Pemetrexed inhibits multiple enzymes involved in folate metabolism and DNA synthesis. This combination has been studied primarily for the treatment of non-small-cell lung cancer (NSCLC).
