bosutinib + fulvestrant + palbociclib

This is a combination therapy being investigated for hormone receptor-positive (HR+), HER2-negative advanced or metastatic breast cancer. The combination targets multiple pathways involved in endocrine resistance: - Palbociclib is a CDK4/6 inhibitor that blocks cell cycle progression from G1 to S phase - Fulvestrant is an estrogen receptor antagonist (antiestrogen therapy) - Bosutinib is a tyrosine kinase inhibitor targeting the Src/Abl pathway, which has been shown to mediate endocrine resistance in breast cancer Preclinical data suggests that adding bosutinib to a CDK4/6 inhibitor and antiestrogen therapy has potential to reverse endocrine resistance in breast cancer.
