The combination of bosutinib and ropeginterferon alfa-2b is an investigational therapeutic regimen being evaluated for the frontline treatment of newly diagnosed chronic myeloid leukemia (CML). This combination therapy, currently under investigation in the Phase 2 BOSURE trial sponsored by St. Olavs Hospital, pairs bosutinib, a second-generation tyrosine kinase inhibitor (TKI), with ropeginterferon alfa-2b, a long-acting monopegylated interferon. Bosutinib functions by inhibiting the BCR-ABL1 fusion protein and SRC family kinases, which are critical for the proliferation of leukemic cells. Ropeginterferon alfa-2b acts as an immunomodulator, binding to Type I interferon receptors to stimulate an immune response against leukemic stem cells. The primary objective of this combination is to enhance deep molecular response (DMR) rates and increase the number of patients who can successfully achieve treatment-free remission (TFR).
