Drug intelligence / Profile preview
Botensilimab + balstilimab + agenT-797 is an investigational combination immunotherapy regimen consisting of three distinct agents: botensilimab (an Fc-enhanced IgG1 monoclonal antibody targeting CTLA-4), balstilimab (a fully human IgG4 monoclonal antibody targeting PD-1), and agenT-797 (an allogeneic, off-the-shelf invariant natural killer T [iNKT] cell therapy). Developed by Agenus and its spinout MiNK Therapeutics, this combination is designed to overcome resistance to standard checkpoint inhibition by combining dual checkpoint blockade with innate and adaptive immune activation. The regimen is currently being evaluated in investigator-initiated Phase II clinical trials, including studies for previously treated, PD-1-refractory advanced gastroesophageal cancers (in combination with ramucirumab and paclitaxel) and previously treated microsatellite-stable (pMMR) metastatic colorectal cancer with liver metastases.
