Overview

Botulinum neurotoxin Type A inhibitor is a small-molecule therapeutic being developed by Hawaii Biotech for the treatment of botulism. The drug is designed to target the light chain (LC) of the Botulinum neurotoxin Type A (BoNT/A), which is a zinc-dependent metalloprotease. In a botulism infection, BoNT/A cleaves SNAP-25, a protein essential for the fusion of synaptic vesicles with the presynaptic membrane, thereby blocking the release of acetylcholine and causing flaccid paralysis. By inhibiting the enzymatic activity of the toxin's light chain, this small molecule aims to prevent the cleavage of SNAP-25, halting the progression of paralysis and potentially allowing for faster recovery. This program is part of Hawaii Biotech's focus on developing medical countermeasures for biodefense and infectious diseases.
