Overview

Botulinum toxin alphavaccine is an investigational RNA-based replicon particle vaccine developed by AlphaVax for protection against botulism, primarily for biodefense applications. The vaccine utilizes the Venezuelan Equine Encephalitis (VEE) virus replicon particle (VRP) platform. In this system, the structural protein genes of the VEE virus are replaced with sequences encoding botulinum neurotoxin (BoNT) antigens, typically the non-toxic carboxy-terminal portion of the heavy chain (Hc). Upon administration, these replicon particles infect host cells and undergo a single round of high-level antigen expression without producing new infectious virus particles. This process triggers a robust immune response, including the production of neutralizing antibodies that prevent the toxin from binding to nerve endings, thereby providing immunity against botulinum neurotoxin exposure.
